Review the Design

1. Select Review Design to review and order the final design.
2. Review the design details in the Review Design pane at the top of the page.

Depending on the design type selected when configuring the design, some or all of the following information is displayed:

Field

Description

Solution Type

The type of design selected when configuring the design (Full Custom, Add-On, or Fixed).

Content Included

The fixed content panels selected for Add-On or Fixed designs.

Selected Targets

The number of targets added to the design followed by the total number of targets across all regions in the design.

Cumulative Target Length (bp)

The total cumulative length of all nonoverlapping target regions in base pairs.

Overall Content Probes

The total number of probes in the design, including add-on and base content if applicable.

Base Content Probes

The number of probes in the design that are used to cover any base content of an add-on design.

Overlap

Percentage of total base pairs in the current design that are targeted for enrichment by more than one target region.

Number of Probes

The number of probes designed to cover the targets. Directly related to library size, probe interval, and sequence characteristics of the designed targeted regions.

Number of Gaps

The total number of gaps in the design. A gap is any base position in the region of interest (exon or otherwise) that is intended for enrichment, but is not theoretically enriched by the probe set. Gaps are one or more bases long and occur more frequently at the ends of targeted regions when dense spacing is not used.

Total Gap Length (bp)

The cumulative base pair length of all gaps in the design.

Duplicate Targets

The number of duplicate targets in the design.

Average Region Size (bp)

The average continuous region size, in base pairs, targeted by DesignStudio while building the design.

Warnings

Any design warnings are displayed here. The two possible warnings are as follows.

Custom Probe Minimum not reached—The design does not contain the minimum number of required probes. Full Custom designs require a minimum of 2,000 probes. Add-on designs require a minimum of 350 probes.
Duplicate Targets—The design has duplicate targets.

Coverage

The percent of target bases covered by the probes.

3. View detailed information on regions, targets, probes, and gaps in the final design using the appropriate tabs in the table below the Review Design pane:

4. [Optional] Select Edit Design to return to the Edit Targets page and manage targets in the design. See Manage and Export Targets.
5. To export information about targets, select the checkboxes next to the appropriate targets, and then select Export.
6. To generate a file suitable for importing into DesignStudio projects:
  1. Select a file format under Save targets as a Coordinate import file:
  2. .csv
  3. .tsv
  4. Select Export.
7. To export more specific information from the design:
  1. Select the type of data under Export data from:
  2. Region
  3. Target
  4. Amplicon
  5. Gap
  6. Select a file format under Save data as:
  7. .csv
  8. .tsv
  9. .bed
  10. Select Export.
8. When ready to place an order, select Get Pricing.

If there are undesigned probes in the final design or an insufficient number of attempted probes selected, the Order button is disabled.

9. Review the final design details, and then select Finalize Design & Get Pricing.
